"Somewhere in time" with Christopher Reeve is kind of like that. Its not that old - it came out in 1980. Also, the woman isn't Audrey Hepburn - its Jane Seymour. He sees a picture of a woman (Seymour) and is instantly drawn to it. But its not like the portrait comes to life - it has more to do with time traveling - its kind of a complicated story.
I am not sure if that is the movie you are thinking of, but it is a really good, romantic movie! http://www.imdb.com/title/tt0081534/

Since it's an old movie you're talking about I think you might be referring to "Laura" (I think that's the name of the title). I believe Humphrey Bogart is in it. It's about a detective and his fascination with "Laura."(I believe she is missing or thought to be murdered). There is a portrait of Laura on the wall and Laura is played by Lauren Becall sp? Didn't like the movie, so I can't remember it very well!

A mysterious girl inspires a struggling artist.
"PORTRAIT OF JENNIE" Starring Jennifer Jones (not Audrey Hepburn) Also Joseph Cotten, Ethel Barrymore, Lillian Gish And Cecil Kellaway. (1948)
