I’m not sure that the movie states this, but it likely does. Some are of the belief that in 2012 the earth will shift magnetic polarity (the poles will reverse, and a compass will point south instead of north, as it does currently). This shift is what is responsible for the calamity in the movie.
Edit: You were asking about the ‘physical’ cause, weren’t you? Others are citing the Mayan calendar, etc. The Mayan calendar was/is incredibly accurate given what was known at the time of its creation. It stops abruptly on Dec. 21 (some claim the 12th), 2012. There are also those who contend that all of the world events currently taking place are lining up for the confirmation of the revelation in the bible. Some Muslims also believe that things are currently staged for the return of the Twelfth Imam.
But from a physical standpoint, the reversing magnetism of the poles would result in all of the tectonic plates shifting. If the tectonic plates shift dramatically, it would cause a lot of natural disasters, such as earthquakes, tsunamis, volcanic erruptions and the like. These plates interact now, causing these events.
Look around you, the movie 2012 may be fiction, but it wouldn’t be the first time a fictional book has predicted the future!! If we, the human race, don’t get our act together pretty soon, it may very well happen!! It will only take ONE missle to set the ball rolling, by mistake or by intention, the end result will be the same!!! To answer your question as to what causes this cataclysim – People!

According to the trailer and other viral videos, it will be a compilation of natural disasters. It actually apparently gets really deep into the conspiracy.
It’s a sci-fi disaster movie with more fiction than science. They were popular in the 50s-70s. The movie is just a remake of those kinds of movies.
